COMBINED EFFECT OF HIGH-ENERGY FOTON RADIATION AND ETOPOSIDE ON PROAPOPTOSIS SPHINGOLIPIDE SYNTHESIS IN GUERIN’S CARCINOMA

Combined effect of high-energy photon radiation and etoposide on sphingolipide synthesis (ceramide and sphingosin), apoptosis inductors, in Guerin’s carcinoma was studied. Tumor was irradiated at Clinac 600, linear accelerator, at two 5 Gr fractions with 24 hours intervals between sessions; etoposide was introduced at the dose of 8 mg/kg abdominally 24 hours before the first irradiation session. It was found that under combined action of irradiation and etoposide in Guerin’s tumor, proapoptosis sphingolipide synthesis was significantly increased (ceramide by 180 %, sphingosin by 143 %) as compared to intact control. The combined effects of high-energy photon radiation and etoposide of potentiation in ceremide synthesis and synergy in sphingosine formation direction were found for the first time. Obtained results show the possibility of radiation modification usage for induction of ceramide way apoptosis and open prospects of searching for new ways of tumor radiation response control.
